چکیده

The article considers the production of red blood cell “ghosts” and their application as a model for studying Na+, K+ - ATPase. Various variants liposomes are used test objects ion transport, but best biological membranes transport ATPase pumps «ghosts» cells. activity Na+/K+ is determined mainly in suspension “ghosts”. This due to need remove interfering compound – hemoglobin obtain that necessary accurate measurement enzyme. In this scientific work, studies were conducted determine concentrations Na+ ions cells cows calves various ages by atomic absorption emission spectroscopy. values these “Ghosts” with set concentration sodium potassium obtained method J. Only solid red blood cell ghosts transport K+, and Na+ against concentration gradients: hollow intact ghosts with K+-Na+ activated ATPase do not.

Human red blood cell ghosts, prepared by simple hypotonic lysis are filled with cytoplasmic proteins. These solid ghosts can transport K+ and Na+ against concentration gradients in the presence of ATP. In hollow red cell ghosts prepared by repeated hypotonic lysis followed by high salt wash, this ability to transport K+ and Na+ in the presence of ATP is lost even though these hollow ghost membr...

Electrophoretic Separation of Different Phophosproteins Associated with Ca-ATPase and Na,K-ATPase in Human Red Cell Ghosts

Ca has been found to increase the quantity of (32)P incorporated into red cell ghosts from [gamma-(32)P]ATP over the levels obtained by incubation with Mg alone or with Mg + Na, in correlation with the effect of Ca on the associated ATPase activities. When the (32)P-labeled ghosts were solubilized in sodium dodecyl sulfate (SDS) and electrophoresed on acrylamide gels only two bands could be det...

[3H]Ouabain binding and Na+, K+-ATPase in resealed human red cell ghosts

The interaction of the cardiac glycoside [3H]ouabain with the Na+, K+ pump of resealed human erythrocyte ghosts was investigated. Binding of [3H]ouabain to high intracellular Na+ ghosts was studied in high extracellular Na+ media, a condition determined to produce maximal ouabain binding rates. Simultaneous examination of both the number of ouabain molecules bound per ghost and the correspondin...

Na-K-dependent ATPase in red cells and thyroid status.

The Relationship between ouabain-sensitive ATPase (Na-K ATPase) activity in erythrocytes and the thyroid status was studied in 36 patients with Graves' disease and 58 patients receiving L-thyroxine (T4) replacement therapy. Forty normal children served as control. Total ATPase activity in 4 untreated hypothyroid patients was significantly reduced (11.0 +/- 4.6 vs 17.3 +/- 4.1 micrograms-P/h/mg-...

Quantitative relationships between the concentration of proteins and the concentration of K+ and Na+ in red cell ghosts.

The concentration of K+ accumulated in and Na+ extruded from red blood cell ghosts are quantitatively correlated with the total protein contents of the ghosts with a linear correlation coefficient of +0.80. The straight line derived from the 43 sets of data on K+ accumulated in the presence of varying ghost proteins, when extrapolated to the protein content of a normal red blood cell, yields a ...
